Peroxisomes Extend Peroxules in a Fast Response to Stress via a Reactive Oxygen Species-Mediated Induction of the Peroxin PEX11a.

Peroxisome is an organelle that is very dynamic and is metabolically active which play important roles in cell function, including reactive oxygen species (ROS) metabolism. peroxisomal dynamics, such as proliferation, movement, and dynamic extension called peroxules production, has been associated with ROS in plant cells.

However, the function and regulation of peroxules largely unknown. Using confocal microscopy, we have shown that treatment of Arabidopsis leaf with heavy metal cadmium produces a time-dependent course peroxisomal dynamics change, starting with the formation of peroxule, followed by peroxisome proliferation, and finally back to normal morphology and number. These changes are for the treatment of Cd regulated by NADPH oxidase (C and F) -related ROS production.

Peroxule formation is a common response to stimuli such as arsenic and regulated by peroxin 11a (PEX11a), as pex11a RNAi Arabidopsis lines can not produce peroxules under stressful conditions. Pex11a line showed higher levels of lipid peroxidation content and lower expression of genes involved in antioxidant defense and a signal, indicating that this extension is involved in regulating ROS accumulation and ROS-dependent gene expression in response to stress.

Our results indicate that the formation peroxule PEX11a and play a key role in organizing the perception of stress and rapid cell response to environmental cues.

Mutations in the gene-coding peroxin contribute PEX22 high malate production in Saccharomyces cerevisiae.

Saccharomyces cerevisiae produce organic acids such as succinic, acetic and malic during alcoholic fermentation. Since contribute malate with a pleasant taste of sake (Japanese alcoholic drink), a variety of methods for breeding yeast strains that produce high-malate has been developed. Here, the high-malate-producing yeast strain isolated F-701H. This mutation, which appears to cause dysfunction Pex22p, sufficient to confer increased productivity and sensitivity DMS malate into the yeast cells. Next, we investigated the mechanism of this mutation causes production of malic high in yeast cells. Peroxins, like Pex22p, retain peroxisomal biogenesis. Analysis of 29 disruptants PEX revealed increased production of malic by the removal of the gene encoding the peroxins responsible for importing proteins (containing peroxisomal targeting signal 1, PTS1) into the peroxisomal matrix, and those responsible for the assembly peroxins in peroxisomal membrane.

A defect in peroxisomal malate dehydrogenase (Mdh3p), save endogenous PTS1, inhibiting malate-producing phenotypes high in the mutant PEX22. Additionally, Mdh3p, which usually sorted into peroxisomal matrix, it has the potential to mislocalized to the cytosol in the mutant PEX22. This suggests that the increased production of malic generated from mislocalization of Mdh3p of peroxisome into the cytoplasm because of the loss of peroxin-mediated transport. Thus, this study reveals a new mechanism for the production of organic acids in yeast for sake brewing.
