Localization of a Trypanosome Peroxin to the Endoplasmic Reticulum.

Trypanosoma brucei is the causative agent of diseases that affect 30,000-50,000 people annually. Trypanosoma brucei harbor unique organelles called glycosomes essential for the survival of the parasite, which requires growth under fluctuating environmental conditions.

The mechanisms that regulate biogenesis of these organelles are poorly understood. Glycosomes evolutionary related to the peroxisomes, which can proliferate de novo from the endoplasmic reticulum or through growth and division organelles that exist depending on the organism and environmental conditions.

Environmental influences on biogenesis glycosome unknown. Here, we show that the membrane protein glycosome, TbPex13.1, localized to glycosomes when cells are cultured under high glucose conditions and the endoplasmic reticulum in low glucose conditions. Low glucose localization is dependent on the presence of the C-terminal tripeptide sequence. Our findings indicate that the biogenesis glycosome affected by extracellular glucose levels and adds to the evidence that de novo biogenesis glycosome occur in trypanosomes.

Since the motion peroxisomal membrane protein is a hallmark of ER-dependent peroxisome biogenesis, TbPex13.1 may be a useful marker for the study of such processes in trypanosomes.

Identification and characterization of genes MoPEX1 peroxin 1 is required for morphogenesis-related infection and pathogenicity in Magnaporthe oryzae.

Peroxisomes required for pathogenicity in many phytopathogenic fungi, but the relationship between the pathogenicity of the fungus and peroxisomal function is not fully understood. Here, we report the identification of T-DNA insertional mutants of Magnaporthe oryzae C445, which was damaged in pathogenicity.

Mutation analysis confirmed the insertion into MoPEX1 gene, which encodes a homolog alleged to peroxin 1. Targeted gene deletion mutants MoPEX1 pathogenic and disturbances in vegetative growth, conidiation and appresorium formation. ΔMopex1 mutant form is not normal, less pigmented, and appresoria nonfunctional, but they were not able to penetrate the plant cuticle. ΔMopex1 mutant is defective in the utilization of fatty acids (eg, olive oil and Tween-20). In addition, the removal MoPEX1 mobilization and degradation significantly impaired lipid droplets during appresorium development. Interestingly, deletion MoPEX1 blocked peroxisomal matrix protein import.

Analysis of M. oryzae strain expressing GFP-fusion MoPEX1 and RFP-PTS1 MoPex1 revealed that localizes to peroxisomes. Yeast two hybrid experiments showed MoPex1 that physically interact with MoPex6, peroxisomal matrix protein essential for fungal morphogenesis and pathogenicity. Taken together, we conclude that MoPEX1 play an important role in peroxisomal function and necessary for infection-related morphogenesis and pathogenicity in M. oryzae.

Functional areas of peroxin Pex19 required for peroxisome biogenesis.

The peroxins Pex19 and Pex3 plays an indispensable role in membrane proteins (PMP) peroxisomal biogenesis, peroxisome division and inheritance. Pex19 play some role in this process, but how these functions are related to the structural organization of the domain Pex19 is unresolved. For this purpose, using mutant deletions, we mapped the areas Pex19 required for peroxisome biogenesis in yeast Pichia pastoris Surprisingly, peroxisomes import competent still formed when Pex19 domain previously thought to be required for the biogenesis deleted, although the size of the peroxisome is greater than that in the cell type wild. In addition, these mutants exhibited a delay of 14-24 hours in peroxisome biogenesis.
