Peroxin 16 (PEX16) plays an important role in peroxisomal membrane protein targeting and de novo biogenesis of the peroxisomes from the endoplasmic reticulum (ER).
We identify trypanosomal PEX16 based on characteristics of a particular sequence and indicates that it is an integral membrane protein glycosomal of procyclic and bloodstream trypanosomes form. RNAi-mediated knockdown partial PEX16 Trypanosoma brucei bloodstream form trypanosomes causing severe ATP depletion, disability motility and cell death. Microscopic and biochemical analyzes revealed a drastic reduction glycosome and mislocalization amount of enzyme matrix glycosomal to the cytosol. The results showed that PEX16 trypanosomal very important for the biogenesis glycosome and thus, providing a potential drug target for sleeping sickness and related diseases.
Peroxin Pex14p are key components for autophagic degradation of mammalian peroxisomes coordinated by directly binding LC3-II.
Pexophagy can be experimentally induced in mammalian cells by removing serum culture. Pex14p, an important membrane protein peroxisomal for imported protein matrix in docking receptor soluble Pex5p, is involved in the degradation of autophagic mammals from peroxisomes and interact with forms lipidated of LC3, called LC3-II, is an important factor for the formation of autophagosome, under conditions of starvation in cell CHO-K1. However, the molecular mechanisms underlying the interaction Pex14p-LC3-II is still unknown.
To verify whether Pex14p directly bind LC3-II, we reconstituted in vitro system of conjugation for the synthesis of LC3-II. We show here that Pex14p directly interact with LC3-II through the transmembrane domain of Pex14p. Pex5p competitively inhibits this interaction, implying that Pex14p preferential binding to Pex5p under rich conditions nutrisi-. In addition, mutants defective in the import Pex5p PTS1-protein loses its affinity for Pex14p under conditions of malnutrition, so it might explain why Pex14p preferentially interact with LC3-II under starvation conditions in vivo.
